Composition:
1 tabl contains 300 mg or 500 mg of paracetamol.
Action:
The drug with analgesic and antipyretic effect, mainly by inhibiting the activity of prostaglandin cyclooxygenase at o.u.n. Well absorbed from the gastrointestinal tract, the maximum concentration in the blood reaches after about 30-60 min, penetrates into body fluids and internal organs, in about 25% bound to plasma proteins. T0,5 is 3-4 hours. It is metabolized in the liver, inducing microsomal enzymes there. It is excreted in the urine in the form of glucuronides and sulphates and after conjugation with cysteine. In patients with renal failure, conjugated Paracetamol accumulates in the blood. After high doses, N-acetyl-p-benzoquinone in the body is probably responsible for causing liver damage.
Contraindications:
Hypersensitivity to the components of the preparation. Alcoholic disease. Severe hepatic or renal failure.
Precautions:
Caution in patients with impaired renal and / or liver function and in the case of glucose-6-phosphate dehydrogenase deficiency. Risk of liver damage in people who are starved and regularly drink alcohol.
Pregnancy and lactation:
The preparation can only be used in extreme necessity.
Side effects:
Rare: hypersensitivity reactions (eg rash, urticaria, rashes). Several cases with typical symptoms of a startling shock. Very rare: nausea, vomiting, digestive disorders, thrombocytopenia, methemoglobinemia, leukopenia, pancytopenia, agranulocytosis, occasional haemolysis in people with blood cell enzymopathy, deficiency of glucose-6-phosphate dehydrogenase mainly found in the Mediterranean and Black people.
Dosage:
Orally. Adults and children over 12 years of age: 1-2 tabl. 3-4 times a day at intervals of not less than 4 hours. Children aged 6-12: 1/2 -1 tabl. 3-4 times daily at intervals of not less than 6 hours. In case of renal failure, the dose should be reduced or the interval between dosing should be extended to 6 hours (adults). The maximum daily dose of paracetamol is 4 g - 8 tablets. (500 mg each) or 13 tablets. (300 mg each) and correspondingly less in those who, for various reasons, have prolonged the period between doses.
