1 suppository contains 80 mg, 150 mg or 300 mg of paracetamol.
Action:
The drug with analgesic and antipyretic effect, mainly by inhibiting the activity of prostaglandin cyclooxygenase at o.u.n. It is well absorbed from the gastrointestinal tract, reaches the maximum concentration in the blood after about 20-60 min., Penetrates into body fluids and internal organs, ca. 25% bound to plasma proteins. T0,5 is 2-3 hours. It is metabolized in the liver, induces microsomal enzymes there; excreted in the urine in the form of glucuronides and sulphates and after conjugation with cysteine. In patients with renal failure, conjugated Paracetamol accumulates in the blood. After high doses, N-acetyl-p-benzoquinone in the body is probably responsible for causing liver damage.
Contraindications:
Hypersensitivity to the drug (rarely seen), congenital G-6-PD deficiency and methemoglobin reductase. Caution should be exercised in patients with bronchial asthma allergic to Acetylsalicylic acid and when repeated doses are used in patients with severe hepatic impairment. It should not be used in children under 3 months of age.
Precautions:
Pregnancy and lactation:
Side effects:
There are isolated cases of hematological disorders with long-term use (thrombocytopenia with symptoms of thrombocytopenic purpura) and cutaneous allergic reactions (urticaria, erythema, pruritus, rash). When used in high doses or for long periods of time, it may impair liver function.
Dosage:
In children under 3 years of age paracetamol can be used only on the express request of a physician. Recommended rectal dose for children over 3 months of age: 10-15 mg / kg m.c. 4 times a day, in selected situations every 4 hours - this dose can be used up to 3 days. Do not exceed the daily dose of 60 mg / kg m.c. Adults and adolescents over 15 years of age 600 mg 1-4 times daily. Do not exceed the dose of 3.0 g per day. There should be a gap of at least 4 hours between doses. For severe renal insufficiency (creatinine clearance less than 10 ml / min), the dose interval should be at least 8 hours.
