The drug acts as an analgesic in headaches, throats, migraines, toothaches, bone, joint and muscle pain and dysmenorrhea. In addition, it can be used in colds and flu-like conditions. It works antipyretic.
Composition:
1 tabl contains 500 mg of paracetamol.
Action:
Analgesic and antipyretic. Inhibits the biosynthesis of prostaglandins by inhibiting arachidonic acid cyclooxygenase at o.u.n. Only slightly inhibits peripheral cyclo-oxygenase enzyme, without causing damage to the gastric mucosa. It does not affect platelet aggregation. Paracetamol is quickly and almost completely absorbed from the gastrointestinal tract and is evenly distributed to body fluids. It binds to small plasma proteins. It is metabolized in the liver and almost completely excreted in the urine in the form of a conjugated - glucuronide and sulfate. A small amount (approximately 5%) of the potentially hepatotoxic intermediate metabolite N-acetyl-p-benzoquinone (NAPQI) is completely conjugated to glutathione and excreted in combination with cysteine or mercapturic acid. If high doses of paracetamol are used, hepatic glutathione stores may be depleted, resulting in the accumulation of a toxic metabolite in the liver. This can lead to hepatocyte damage, necrosis and acute liver failure. Less than 5% of the paracetamol dose is excreted unchanged. Medium T0,5 is 1-4 hours.
Contraindications:
Hypersensitivity to paracetamol or to any of the excipients. Alcoholic disease. Severe hepatic or renal failure.
Precautions:
Do not take other medicines containing paracetamol. To be used with caution in patients with G-6-PD deficiency and methemoglobin reductase. Patients with liver or kidney failure should consult a physician before using the medicine. Do not drink alcohol while taking the drug due to the increased risk of liver damage. The special risk of liver damage exists in patients who are starved and regularly drink alcohol.
Pregnancy and lactation:
Epidemiological studies conducted in pregnant women did not show any adverse effects of paracetamol administered at the recommended doses. Paracetamol is excreted in breast milk in amounts that are not clinically relevant. Available published data do not contain contraindications to breastfeeding while taking the drug. Like other medicines, the preparation should be used during pregnancy and breastfeeding if necessary.
Side effects:
Very rare: thrombocytopenia, anaphylactic reactions, skin hypersensitivity reactions including skin rash, angioneurotic edema and Stevens-Johnson syndrome (bladder erythema multiforme), bronchospasm in patients with hypersensitivity to Acetylsalicylic acid and other NSAIDs, liver dysfunction.
Dosage:
Orally. Adults and children over 12 years: 2 tablets; if necessary, the dose can be repeated no more frequently than every 4 hours, up to 4 times a day. The maximum daily dose: 4000 mg (8 tablets). Children aged 6-12: 1/2 - 1 tabl; if necessary, the dose can be repeated no more frequently than every 4 hours, up to 4 times a day. The maximum daily dose: 60 mg / kg / 24 h administered in divided doses 10-15 mg / kg You should not use it regularly for more than 3 days without consulting your doctor. Tablet is not recommended for children under 6 years of age - use a suspension.
