Composition:
1 tabl contains 5 mg or 10 mg of vinpocetine. The tablets contain lactose.
Action:
Vinpocetine has a positive effect on the metabolism of brain tissue by improving microcirculation. It increases the consumption of Glucose and oxygen, and thus the production of ATP. Increases the tolerance of nerve cells to hypoxia. Vinpocetine affects the transmission of noradrenaline and serotonin. By inhibiting phosphodiesterases in vascular wall cells, it increases cAMP and cGMP concentrations, resulting in smooth muscle relaxation and vasodilatation. It improves microcirculation in the brain without creating a stealth effect. Inhibits dose-dependent sodium channels, thereby exhibiting neuroprotective and anticonvulsant effects. The drug is well absorbed from the gastrointestinal tract (approximately 60%), the presence of nutritional content increases the absorption, Cmax in the blood is obtained after 0.8-1.5 hours, biological T0,5 is 4.83 ± 1.29 h. In the body, the drug undergoes metabolism (mainly in the liver) to apovincaminin, excreted mainly in the form of free urinary (60%) and faecal (40%).
Contraindications:
Hypersensitivity to the active substance or to any of the excipients. Finding or suspicion of intracerebral bleeding (acute phase of haemorrhagic stroke). Heart arythmia. Diagnosis of ischemic heart disease. Pregnant women planning pregnancy and breastfeeding women. Children.
Precautions:
Before starting treatment, other causes of brain dysfunction should be excluded. Use with caution in patients with hypotension or orthostatic hypotension. ECG monitoring is recommended in patients with long QT syndrome or using an additional drug that causes QT prolongation. Due to the lactose content, the drug should not be used in patients with galactose intolerance, lactase deficiency (Lapp type) or malabsorption of glucose-galactose.
Pregnancy and lactation:
Use in pregnant women, planning pregnancy and breast-feeding is contraindicated.
Side effects:
Table. 5 mg. Uncommon: tachycardia or extra heart spasms, ECG changes (ST segment depression, QT prolongation), low blood pressure. Rarely: heartburn, abdominal pain, nausea, insomnia, pain and dizziness, weakness. Very rare: cutaneous allergic reactions, decrease in the number of white blood cells, reduction of hematocrit and hemoglobin level in the peripheral blood, increase in liver enzymes.Table. 10 mg. Uncommon: sleep disorders (insomnia, drowsiness), dizziness and headache, weakness (these symptoms may be related to the underlying disease), changes in blood pressure (mainly lowering of blood pressure), headache, nausea, heartburn and dry mouth oral, increased liver enzymes, allergic skin reactions, ST segment depression, QT prolongation, tachycardia and additional heart contraction. Rarely: hypotension, abdominal pain, leukopenia. Very rare: decrease of hematocrit and hemoglobin level in peripheral blood.
Dosage:
Orally. Adults: initially 5-10 mg 3 times a day; Maintenance dose: 5 mg 3 times a day. No dosage adjustment is necessary for patients with impaired hepatic and / or renal function. The drug should be administered regularly.
